Nucleoside-modified mRNA technologies necessarily incorporate N1-methylpseudouridine into the mRNA molecules to prevent over-stimulation of cytoplasmic RNA sensors. Despite this modification, mRNA concentrations remain mostly determined through measurement of UV absorbance at 260 nm wavelength (A260).
