Abstract: The hedgehog (HH) gene was first identified in Drosophila as a regulator of larval segmentation7, after which three mammalian homologs were discovered: desert hedgehog (Dhh), Indian hedgehog (Ihh), and sonic hedgehog (Shh)8-11. Shh is the most widely expressed HH ligand and is found in the epithelium and at epithelial-mesenchymal boundaries of various tissues, including the tooth, gut, lung, and limb, where it controls morphogenesis and adult homeostasis12. Precise regulation of Shh signaling is therefore critical for proper tissue development and patterning. Perturbations to the pathway have been linked to severe congenital abnormalities, including polydactyly and holoprosencephaly12.
